History: From UBC to Cell Culture Company
The Rho 1D4 monoclonal antibody was developed by Dr. Robert Molday at the University of British Columbia. Originally developed to recognize rhodopsin, it has been widely used in research involving rhodopsin and recombinant proteins containing the 1D4 epitope tag. Rho 1D4 was previously sold through FlintBox and is now supplied directly by Cell Culture Company.

1D4 Epitope Tag Performance
| Parameter | Data |
|---|---|
| Tag sequence | TETSQVAPA (9 amino acids) |
| C-terminal tag binding | ~10× stronger than N-terminal |
| N-terminal tag binding | Possible; ~10× weaker than C-terminal |
| Calcium dependence | None (calcium-independent) |
| Expression systems | Mammalian and yeast systems, including HEK 293, COS-1, and COS-9 cells |
| Expression vectors used | pcDNA3.1, pCEP4 |
| Elution (affinity purification) | Excess epitope peptide under mild conditions |
Applications
| Application | Use |
|---|---|
| Western blotting | Detect rhodopsin and 1D4-tagged proteins |
| Immunofluorescence / ICC | Localize targets by conventional or confocal microscopy |
| Immunogold labeling | Detect proteins by electron microscopy |
| IP / co-IP | Isolate tagged proteins; study protein-protein interactions |
| Affinity purification | Purify recombinant 1D4-tagged proteins |
Affinity Purification with Rho 1D4
Rho 1D4 can be covalently coupled to affinity matrices using cyanogen bromide or other conventional immobilization linkers. Bound 1D4-tagged proteins can be selectively eluted using an excess of the corresponding epitope peptide under mild conditions.
Storage and Handling
| Condition | Recommendation |
|---|---|
| Short-term | 2–8 °C |
| Long-term | Aliquot; −80 °C with 10% glycerol |
| Freeze-thaw | Avoid repeated cycles; use single-use aliquots |
| Visual check | Visible precipitate may indicate deterioration |
