Stable Cell Line – Service Workflow

We can make our constructs to over-express any genes (wildtype, mutant or synthetic) and to silence/knockout any genes from any species if you don’t have any starting material. Our scientists are available to discuss your specific requirement and come up with the recommended constructs for your specific needed. We can then produce the final construct based on the agreed specification and full-length validated by Next Generation Sequencing as QC.

All clonal stable cell lines are developed based on individual client specifications, using highly efficient gene delivery technologies to produce clonal cell lines. We can produce multiple clonal cell lines depending on your needs, with various expression levels (low / medium / high expression), reprogrammed and genetically modified. Typically, antibiotic resistance or fluorescent reporter gene markers are incorporated into the plasmid DNA construct to facilitate selection process. After we have determined the cleavage efficiency of the pooled cell population, isolate single cell clones for further validation and banking. We isolate single cell clones from the selected pool using limiting dilution cloning (LDC) in 96‑well plates.

We apply a variety of techniques to characterize genetically engineered cell lines to physiologically and molecularly evaluate the modification in the final cell line.

Techniques such as: Immunofluorescence for a qualitative analysis and flow cytometry of quantitative population within the final cell lines.

Moreover, the genomic integration of the target gene and associated mRNA expression increase can be verified using quantitative real-time RT-PCR.  The Western blot analytical technique uses antibodies specific to the target protein in order to quantify the expression of the protein in a sample. The utilization of both techniques validates that the target gene is both integrated into the genome and functionally expressing.

We create a master cell bank with at least 10 passages of 3 different clones. We send vials of each passage and keep vials safe in the bank for an extra banking charge in case you need extra vials for security. The shipment will include a Cell pool/ clone generation report and we will follow up with any step of you downstream processes to ensure the success of your project.
